I was just wondering with the e-manage ultimate if your using the pressure sensor that you buy do you need to use a maf sensor.

No, when you switch over from the MAP sensor from the MAF sensor, you can remove your MAF sensor. I wouldn't recommend doing the switchover until you get to the dyno though because there could be unforseen complications with changing it over that would be addressed during the tuning process.
